The classifying of the four criteria in minority groups are the race, ethnicity, religion, and the gender. Then the groups are racial groups, ethnic groups, religious groups, gender and other groups. The last you have is the subordinate groups that could be any groups they want. The racial group is a group that is a socially set apart because of the obvious physical differences in each other. The ethnic group is a group that can or set apart from others because of its national origin or cultural patterns that can be distinctive. The religion groups is just apart what it can say in its group made up with people that are in the same religion. Gender groups are made up of the male or all female. Now you have the subordinate group that can have a whole different set of criteria such as age, disability status, physical appearance, and the sexual orientation. Then I would say that you have groups that are made up of two or more groups put together, such as religion and color, or another combination that might be gender or sexual orientation, and it could be race. It can or it is usually the people that meet more than one of the groups into this that have the way of hardest times of dealing with things in the country or something that comes to mind. I would say that in this country that people are always placing people in groups that is hard enough to be just one group but when you add other groups into it there are even bigger problems or issues. Then to be a female in this country in these days it can be very hard but if you are one then you can add the race into it. The last part is religion to it and the people are more likely to feel something totally different about you. When we said this is all done we the people put too much importance on what groups that we belong to. I do believe and think that people put labels on the people because they are down deep what they want to feel the superior to all others. The best part in some subconscious ways we put people in these groups and place labels on them to help other boost their egos and make them feel better about themselves. The labeling and the grouping people for hundreds of years and this is not a new thing by any means at all. This has been all the way for people to keep other down were they thought they belonged to. By this way in which people try and make themselves feel better about themselves for the simple fact that can do so many to the people that would have low self-esteem, and it is the way of boosting their own confidence. It is said realty of the way of life that if the people are the type to place people in groups they will not have anything to do with anyone else but for those that are in the same groups as they are in. I would say that a lot of people only have friends from people in which are inside their own group of people. But I do also think if we stop grouping people and putting labels on them we would have such better world. Look at all the problems down through our history that all that happened because of the labeling people. I might have an example of this such as Hitler he place people in groups and also label them and put them into separate camps. Then he thought unless you were a blonde hair and blue eyed you were not good enough to live in his world. Plus he thought the only way to deal with any problems that this could cause so he had an ethnic cleansing by killing those people and we all know how it turn out in that time and day. But he was is far from the only one that has done that. During that same time when the World War 2 was going own and look at what happened in our very own country we placed Japanese American’s in camps for the fact they were Japanese And still to this day and time we separate people into so many groups and keep thinking that are we all not just part of the human race or do we try to something else all the time.

Asses the role of Ulysses S. Grant in the victory of the union Ulysses S. Grant played a crucial role in the victory of the union in the American civil war. Ulysses S. Grant, later to become the 18th president of the United States, commanded union forces during the civil war as a general and general in chief. The greatest assets that Ulysses S. In a battle that was to become the bloodiest up to that date in American history, with 23,000 casualties, Grant refused to retreat, he eventually managed to stabilize his line and with the help of reinforcements produced a counterattack and turned a severe loss into a victory. Grant received heavy criticism for his decisions at Shiloh but Lincoln defended him stating "I can't spare this man, he fights" One of Grants greatest military success of the war was the siege of Vicksburg, July-August 1863. Grant spent the winter of 62-63 attempting and failing multiple operations to gain access to the town, Grant crossed the Mississippi and headed for Jackson, he took a great risk and cut away from his supplies to sever the railroad to Vicksburg effectively cutting the confederate garrison off from reinforcements. Grant now defeated the confederate army at Champion Hill and then pursued the retreating army to the garrison at Vicksburg. The six week siege began. On the 4th of July 1863, being cut off and with no possibility of relief confederate general John Pemberton surrendered. This union victory in conjunction with the battle of Gettysburg the previous day is considered the turning point in the war.

If the companies were to go to court, deciding in where the court hearing would take place is another issue. If the hearing was to be held in the foreign country, the laws might not be the same and that country might not have much experience in international disputes. If the hearing was to be held in the United States, the other company and the country’s government it resides in could become hostile and not uphold the ruling. Arbitration would be the best way to resolve any disputes between the two countries. Arbitration can save a lot of time and money for both companies while also protecting the images of both companies by not having a lengthy trial in the press. â€Å"Arbitration is often less costly, less litigious, and less time-consuming, and offers more privacy to the parties than litigation† (Companies Export, 2008).The ruling from the arbitration could be binding or non-binding depending on what is agreed upon in the contract. The U.S. based company needs to be aware of the laws that affect the company and the business dealings in the other country. The first consists of laws, such as antitrust, employment, and economic-espionage laws, that are also applied frequently in the domestic context. With respect to these laws, the international business activities of the U.S. companies are a rguably at special risk because management educated and trained outside the United States might be less familiar with these laws than management educated and trained in the United States† (Winer, 2013). Bribery is also a consideration when taking legal action against a foreign business partner because this is illegal under the U.S. Foreign Corrupt Practices Act (FCPA). In some countries, bribery and gift giving is not seen as an illegal or unethical act but a form of showing respect and gratitude. A company needs to be careful on how this is perceived because it could be seen as an illegal act on the company’s part. Even if CadMex was to invest the time and the money into sublicensing the drug, CadMex would lose money because the drug is being sold at a subsidized price. The royalties that would be paid out with sublicensing still would not gap the margin of loss that CadMe x has. When Customs and Laws Conflict When the local customs and the local laws conflict with the customs and laws of an organization operating abroad; the organization should look at the customs and laws in which it is operating under and those should prevail, as long as the company is following the laws in which it is to operate by. When operating abroad, it is important to be aware and sensitive to the customs of that country. Allowing employees to observe religious and cultural holidays will not only keep moral high but also will keep the company within the laws that country has. Knowing the cultural normalities and integrating those into the business model will allow for the employees and members of the community to feel that the business has their interests mind. Being able to comply with the laws of the country in which the business is in while also complying with the laws set by the country, in which the mother company resides, can be a hard line to walk. Complying with the laws of the country in which was agreed upon in the contract at the onset of the partnership would prevail. Making sure to follow the rules of the country that the business is in is important because that is where the business will produce its goods. â€Å"Employees who engage in international business are responsible for knowing and complying with both the laws and regulations of the countries in which the businesses operate and the U.S. law and regulations that apply outside U.S. borders. If U.S. law conflicts with the local customs, or if the local law is more restrictive than U.S. laws or company policies, one must follow the more restrictive law, custom or policy† (Olin, 2012). Resolving Domestic and International Issues Resolving domestic issues can be done through arbitration or through the court system. It can be as simple as suing a party and showing up to court. When trying to resolve an issue domestically, the laws are well known for both parties and the venue in which the issue will be settled is easily accessible to both parties. Resolving an issue internationally is more work involved. The issue has to be resolved through which venue both parties agreed upon when signing the contract. Both companies need to know the laws of that country and travel might be needed. If arbitration was selected, both parties would need to submit all paperwork to a panel of arbitrators and adhere to the judgment in which the panel decides. The OED gives only one spelling for the noun and only one for the adjective: pendant (noun): a jewel, bead, tassel, or the like [that] hangs down as an ornament. pendent (adjective): hanging; suspended from or as from the point of attachment, with the point or end hanging downwards. Of a tree: having branches that hang or droop down. According to these definitions, the following examples demonstrate correct usage: The diamond pendant at her throat glittered in the candlelight. There are a variety of different types of samples in statistics. Each of these samples is named based upon how its members are obtained from the population. List of Sample Types Random sample – Here every member of the population is equally likely to be a member of the sample.  Members are chosen via a random process.Simple random sample – This type of sample is easy to confuse with a random sample as the differences between them are quite subtle. In this type of sample individuals are randomly obtained, and so every individual is equally likely to be chosen.  It is also necessary that every group of n individuals is equally likely of being chosen.Voluntary response sample – Here subjects from the population determine whether they will be members of the sample or not.  This type of sample is not reliable to do meaningful statistical work.Convenience sample - This type of sample is characterized by the selection of easy to obtain members from the population.  Again, this is typically not a worthwhile style for a sampling technique.Systematic sample - A systematic sample is chosen on the basis of an ordered system.Cluster sample â €“ A cluster sample involves using a simple random sample of evident groups that the population contains. Assembly of Functional Cellulolytic Enzymes In the present study, we reported the assembly of functional cellulolytic enzymes using a synthetic, cell-surface engineered yeast consortium. Trichoderma reesei endoglucanase II (EGII) and cellobiohydrolase II (CBHII) and Aspergillus aculeatus ÃŽ ²-glucosidase I (BGLI) were displayed as fusion proteins with the AGA2p C-terminus of a-agglutinin on the cell surface of the diploid yeast strain Saccharomyces. cerevisiae Y5. The immobilization of each enzyme on the cell surface was confirmed by immunofluorescence microscopy. This type of yeast consortium allowed convenient optimization of ethanol production by adjusting the combination ratios of each cell type for inducing synergy in cellulose hydrolysis. Next, the direct ethanol fermentation from steam-exploded corn stover was investigated. The optimized cellulase-displaying consortium produced 20.4 g/l ethanol from 48.4 g cellulose per liter after 72 h in the presence of a small amount of cellulase reagent (0.9 FPU/ml). These findings suggested the feasibility of the cellulase-displaying yeast consortium for simultaneous saccharification and fermentation. Currently, many technological barriers exist with respect to the economical production of ethanol from lignocellulosic biomasses [1]. In the process of hydrolyzing cellulose into soluble sugars, multiple cellulases including endoglucanase (EG), cellobiohydrolase (CBH), and ÃŽ ²-glucosidase (BGL) are required [2]. Consolidated bioprocessing (CBP), which combines enzyme production, hydrolysis, and fermentation in one step, is a promising strategy for effective ethanol production from lignocellulosic materials. Saccharomyces cerevisiae is the traditional microorganism used for ethanol production, but it is unable to utilize cellulosic materials and a saccharification process is required prior to fermentation to produce glucose [3-4]. Numerous attempts have been made to engineer S. cerevisiae strains to express cellulases by cell surface engineering for direct ethanol production from cellulose, and although various bifunctional or trifunctional cellulose-degrading strains have been const ructed, the efficiency of cellulose degradation has not been sufficiently improved [5-9]. It would appear that co-expression of all cellulolytic enzymes in a single cell resulted in relatively low expression levels of cellulases, which may have been due to the heavy metabolic burden and potential jamming of the secretion machinery [6,7,10]. Therefore, in this study, we adapted a new strategy of performing simultaneous saccharification and fermentation with a synthetically engineered yeast consortium having the desired properties of cellulolytic ability and ethanol production to reduce the metabolic burden. The development of a diploid yeast strain is another promising strategy for improving expression levels of heterologous genes and enhancing the fermentation performance of S. cerevisiae. Because diploid strains have better growth ability as well as stress tolerances compared with haploid strains, they are particularly suited for industrial applications. Previously, our group reported on the construction of an à Ã‚ °-agglutinin expression system for genetic immobilization ÃŽ ²-glucosidase I on the cell surface of S. cerevisiae Y5 (Patent No: ZL200810222897.7, CGMCC2660). This diploid robust yeast strain possessed many advantages, such as higher ethanol yield, higher resistance to ethanol, and higher physiological tolerance to inhibitors present in lignocellulosic hydrolysates. Here, we report on our efforts to demonstrate the assembly of functional cellulolytic enzymes using a synthetic yeast consortium. In this study, we demonstrated the feasibility of constructing a novel cell surface engineered diploid yeast consortium for direct ethanol production from phosphoric acid swollen cellulose (PASC) and steam-exploded corn stover (CS), an important step toward direct ethanol production from insoluble cellulosic materials. The strains and plasmids used in this study are summarized in Table S1. Saccharomyces cerevisiae Y5 used for the yeast cell surface display of the cellulolytic enzymes was a newly developed diploid strain in our laboratory. E. coli Top 10 was used as the host strain for recombinant DNA manipulation. T. reesei was purchased from CICC (China Center of Industrial Culture Collection). E. coli transformants were grown in Luria-Bertani medium (1% tryptone, 0.5% yeast extract and 1% NaCl, pH 7.0) supplemented with 100 ug/ml of ampicillin. S. cerevisiae Y5 transformants were selected and maintained on Geneticin plates (1% yeast extract, 2% peptone and 2% glucose supplemented with 600 ug/ml Geneticin) at 30 °C , were induced in YPG (1% yeast extract, 2% peptone, and 2% galactose) at 20 °C. The fermentation medium was composed of 10 g/l yeast extract, 20 g/l polypeptone and 10 g/l PASC as the sole carbon source. The à ¯Ã‚ ¬Ã‚ lamentous fungus T. reesei was cultured in potato dextrose aga r medium (2% potato extract, 2% glucose) at 27 °C. The cDNA was synthesized from mRNA by using the First-Strand cDNA synthesis kit (Fermentas). Unless otherwise indicated, all chemicals, media components and supplements were of analytical grade standard and obtained from Sigma-Aldrich (St. Louis, MO, USA). All restriction enzymes were purchased from New England BioLabs (Ltd. Beijing). Primers used for plasmid construction are provided in Table S2. Plasmid pAGA1 for over-expression of the AGA1 gene and plasmid pBGLI for cell surface display BGLI were constructed previously [11]. Plasmid pEGII for cell surface expression of the EGII (egl2) was constructed as follows. The 1194 bp DNA fragment encoding the egl2 gene without its native secretion signal was amplià ¯Ã‚ ¬Ã‚ ed with the à ¯Ã‚ ¬Ã‚ rst-strand cDNA prepared from T. reesei as the template using primer pairs egl2-For/Rev, this DNA fragment was introduced into the yeast display vector pYD1(Invitrogen) with Kpn I/BamH I. MAT terminator was amplified from pYD1 by using primer pairs MAT-For/Rev and then digested with BamH I/EcoR I to create plasmid pYD1-egl2MAT. The KanR fragment was obtained from plasmid YIP5-KanR by two-step cloning. First, the DNA fragment containing ADH promoter and KanR ORF was amplified from YIP5-KanR by PCR using the KanR-For/Rev primers and inserted into EcoR I/Apa I site of plasmid pYD1-egl2MAT; next, the ADH terminator digested with Bgl II/Nde I was also introduced into pYD1-egl2MAT. The resulting plasmid was named pEGII. For displaying the T. reesei CBHII gene (cbh2) in S. cer evisiae Y5, plasmid pCBHII was created. A 1344 bp gene fragment coding for the mature region of the CBHII was amplià ¯Ã‚ ¬Ã‚ ed using primers cbh2-For/Rev-KT and introduced into plasmid pEGII digested with Kpn I/BamH I for replacing egl2 to form pCBHII (Figure 1). Transformation of S. cerevisiae Y5 was carried out using the lithium acetate method [12]. The plasmid pAGA1 was linearized by Apa I for chromosome integration. The plasmid pYD1 was transformed into S. cerevisiae Y5 as a negative control. S. cerevisiae Y5 clones transformed with different plasmids (strain Y5/pYD1 contained plasmids pAGA1 and pYD1, strain Y5/EGII contained plasmids pAGA1 and pEGII, strain Y5/CBHII contained plasmids pAGA1 and pCBHII) were selected and maintained on Geneticin(G418) plates. Immunofluorescence microscopy was performed as described previously [13]. Immunostaining was performed as follows. Induced recombinant yeast cells expressing cellulases were harvested by centrifugation at 6000 rpm for 5 min and washed with phosphate-buffered saline (PBS). As the primary antibody, mouse anti-Xpress tag antibody (Invitrogen, R910-25) for EGII and CBHII was used at dilution rates of 1:1000. As the second antibody, Fuorescein (FITC)-conjugated goat anti-mouse IgG(H+L) (Jackson, 115-095-003) was used at dilution rate 1:200. Cells and the anti-body were incubated at room temperature. After washing the cell–antibody complex with PBS twice, cellular localizations of the cellulases were observed under a fluorescence microscope. Yeast strains Y5 and Y5/pYD1were used as control. Yeast cells were induced in YPG medium for 48 h at 20 ºC and harvested by centrifugation for 5 min at 6000 rpm, washed with distilled water. BGLI activity of strain Y5/BGLI was measured using à ¯Ã‚ Ã‚ ²-nitrophenyl-ÃŽ ²-D-glucopyranoside as the substrate according to a previously described method [14]. Endoglucanase and cellobiohydrolase activities were determined by hydrolysis of carboxymethyl cellulose (CMC) and phosphoric acid swollen cellulose (PASC), respectively. PASC was prepared from Avicel PH-101 (Fluka Chemie GmbH, Buchs, Switzerland) as amorphous cellulose. The cell pellet was resuspended in a reaction mixture of 1% CMC or 1% PASC in 50 mM sodium acetate buffer (pH 5.0) with the optical density at 600 nm adjusted to 1.0. After a reaction at 50 ºC for 30 min, the activities were determined by DNS method [15]. One unit of enzyme activity was defined as the amount of enzyme released 1 ÃŽ ¼mol reducing sugar from the substrate per minute. The abilities of the engineered yeast consortium (Y5/EGII + Y5/CBHII + Y5/BGLI) to fermentation ethanol from PASC and steam-exploded corn stover were investigated. The steam-exploded corn stover used in this study was provided by Henan Tian Guan Group Co., Ltd (Nanyang, Henan, China). The raw material was chopped to 2-3 cm size and treated in a steam-exploded vessel at 2.0 MPa for 5 min. The pretreated feedstock was dried at room temperature and directly used as a substrate without washing. The moisture content of the substrate was 8%. The composition of materials was quantitatively analyzed following the NREL Laboratory Analytical Procedure NREL/TP-510-42618 (Structural carbohydrates and lignin) (Sluiter et al., 2008)[16], as shown in Table 3. An enzyme mixture composed of equal amounts of cellulase (Sigma-Aldrich, St. Louis, MO) and ÃŽ ²-glucosidase (Sigma-Aldrich) was used. Yeast cells harboring different surface-display plasmid for EGII, CBHII, or BGLI, were grown in YPD medium a nd then transferred to YPG medium for 48 h at 20 ºC to express cellulase. Cells collected by centrifugation at 5000 rpm for 5 min at 4 ºC, washed with distilled water twice, and mixed in the adjustable ratio to a total initial cell concentration of 30 g/l wet weight to form the functional consortium. Ethanol fermentation proceeded at 30 ºC with 90 rpm in 250 ml Erlenmeyer flasks. 1ml samples of the fermentation broth were taken periodically and stored at -4 ºC until they were analyzed for sugar and ethanol content. The total sugar was determined by the phenol-sulfuric acid method [17]. Glucose was measured by HPLC (model 1260, Agilent Technologies) equipped with a Hi-Plex H column 300 mm Ãâ€" 7.7 mm) and a refractory index (RI) detector. Samples were run at a temperature of 60 ºC and a mobile phase of 5 mM sulfuric acid at a flow rate of 0.6 ml/min. Ethanol analysis was carried out using GC (model 7890A, Agilent Technologies) equipped with a flame ionization detector and a HJ-PEG column. Samples were run under the following conditions: column oven at 120 ºC, front injection port at 200 ºC, with N2 as the carrier gas at a flow rate of 4 ml/min. The expression plasmids pEGII and pCBHII (Fig. 1) were transformed into the yeast S.cerevisiae Y5 strains, respectively. All of recombinant yeast strains had a pAGA1 plasmid for integrating AGA1 into the chromosome, and the resultant transformants were designated strains Y5/EGII and Y5/CBHII (Table S1). Upon galactose induction, the proteins were expected to be secreted and interact with the Aga1p and Aga2p anchor system by using the glycosylphosphatidylinositol (GPI) anchor linked to the cell surface. To confirm displaying of EGII and CBHII on the yeast cell surface, immunofluorescence labeling of the cells was carried out using mouse anti-Xpress IgG antibody as the primary antibody. The green fluorescence of Fuorescein (FITC)-conjugated goat anti-mouse IgG was observed for strains Y5/EGII and Y5/CBHII (Fig. 2), indicating that EGII and CBHII were displayed on the cell surface, respectively. The cells harboring the control plasmids were hardly labeled with mouse anti-Xpress IgG(Fig. 2). These results suggested that two types of cellulase were successfully expressed on the cell surface of S. cerevisiae Y5 strain. As shown in Table 1, EGII, CBHII and BGLI activities were detected in the pellet fraction of strain Y5/EGII, Y5/CBHII and Y5/BGLI, respectively. The strain Y5/CBHII and strain Y5/EGII showed moderate CBHII and EGII activity (1.14 U/OD600 and 1.27 U/OD600, respectively). The BGLI activity of strain Y5/BGLI cells was relatively low, which was only 0.72 U/OD600. No enzyme activity was detected in the culture supernatant (data not shown), and the control strain without displayed enzymes exhibited less than 0.1 U/OD600 of enzyme activity. These results clearly indicated that active enzymes were displayed on the cell surface without leakage into the culture medium. Ethanol fermentation from 10 g amorphous cellulose per liter was performed using a cell combination system consisted of three cellulase-displaying yeast populations. Cells displaying EGII, CBHII and BGLI were mixed in various ratios and the produced ethanol from PASC were measured. S.cerevisiae Y5 without displayed enzymes was the control strain. A mixture of cells with EGII: CBHII: BGLI ratio of 2:1:1 produced the highest amount of ethanol (1.76 g/l) after 84 h; the yield (in grams of ethanol produced per gram of consumed reducing sugar) was 0.42 g/g (Fig. 3). A mixture of cells composed of an equal amount of each cell type produced 0.68 g/l ethanol after 84 h (Figure 3), indicating about 1.6-fold improvement of ethanol production by optimizing the cell ratio. However, a large portion of the substrate (the amount of residual sugar after 84 h hydrolysis of 10 g/l PASC was 5.5 g/l, and the sugar consumption rate was 43.3%) remained after 96 h without being hydrolyzed because the cellu lase activities displaying on cell surface were not enough for complete cellulose digestion. Simultaneous saccharification and fermentation of steam-exploded corn stover (CS) as a sole carbon source was conducted for the cellulase-displaying yeast consortium of the optimized ratio 2:1:1 in the presence of commercial cellulase (Sigma-Aldrich, St. Louis, MO) with different enzyme loadings (0, 0.3, 0.6, 0.9, 1.2, 1.5 FPU/ml). A mixture of cells was incubated in 100 ml of YP medium (20 g/l peptone, 10 g/l yeast extract) for 1 h to remove residual carbon source, and then resuspended in YP-CS medium (YP medium containing 100 g/l steam-exploded corn stover, corresponding to 48.4g cellulose per liter). As shown in Fig. 4, in the presence of 0, 0.3, 0.6, 0.9, 1.2 and 1.5 FPU/ml cellulase, 34.49, 18.71, 7.03, 2.11, 1.98, and 1.23 g/l of residual cellulose remained after 84h, respectively. Addition of 0.9 FPU/ml cellulase enabled utilization of 92.3% of the initial cellulose (Figure 4). The cellulose hydrolyzed by cellulase-displaying yeast consortium with an additional 0.9 FPU/ml cellulase was nearly the same as that by control strain S.cerevisiae Y5 with an additional 1.5 FPU/ml. These results indicate that cellulases displayed on the yeast cell surface improve hydrolysis of cellulose, although their activities were lower than commercial enzymes. Furthermore, using the optimized cell combination system, the relationship between the amount of added cellulase and final ethanol concentration was investigated. As shown in Fig. 5, in the presence of 0.9 FPU/ml cellulase, the cellulase-displaying consortium produced 20.4 g/l ethanol after 72 h, which was similar to the value (20.9 g/l) obtained by control strain in the presence of 1.5 FPU/ml cellulase (Table 2). Notably, as the ethanol yield reached 86% of the theoretical yield with 0.9 FPU/ml cellulase, the cell-surface engineered system enabled a reduction in the amount of added commercial cellulase. Hydrolysis of crystalline cellulose to glucose requires the sequential reactions of three groups of cellulases: endoglucanase, cellobiohydrolase, and ÃŽ ²-glucosidase. CBP is a one-step process where all steps occur in a single reactor and a single microorganism or microbial consortium converts pretreated biomass to ethanol with no additional commercial enzymes. The key challenge of CBP lies in choosing the optimal host to directly convert lignocellulosic materials to ethanol. In recent years, several researchers have been engaged in co-displaying multiple cellulases in a single cell for direct conversion of cellulose to ethanol [18-21]. However, the enzyme activity can be limited because of the metabolic burden [22]. Furthermore, it is difficult to control the surface expression level of each enzyme for optimal ethanol fermentation. Apiwatanapiwat et al., constructed the engineered yeast strain NBRC-5Es that co-displayed two types of amylolytic enzymes, two types of cellulolytic enz ymes (T. reesei EGII and CBHII), and A. aculeatus BGLI on the cell surface. The NBRC-5Es strain produced 1.04 g/l ethanol from 8.44 g/l of the acid-treated Avicel after 48 h of fermentation and resulted in a large portion of the substrate remaining without being hydrolyzed by the enzymes. In this study, instead of co-displaying all the enzymes in one cell, we developed a cellulase-displaying yeast consortium consisting of three types of yeast cells, each displaying different cellulases. This method allows for convenient optimization of ethanol production by adjusting the combination ratio of each cell type for inducing a synergy in cellulose hydrolysis. Diploidization is also a promising strategy for enhancing the fermentation ability of S. cerevisiae. Because polyploid yeast strains, including diploid strains, have higher cell growth rates, cell yields, and tolerances to various stresses compared with haploid strains, they are particularly suited for industrial application. Therefore, to generate an efficient â€Å"whole-cell biocatalyst† yeast strain related to cellulosic ethanol production, we selected S. cerevisiae Y5, a robust diploid strain, as the host cell based on its fermentation and inhibitor tolerance properties [23-24]. We first explored the possibility of ethanol fermentation from PASC by using the surface-immobilized yeast consortium (Y5/EGII+Y5/CBHII+Y5/BGLI). A mixture of cells at the optimized EGII: CBHII: BGLI ratio of 2:1:1 produced 1.6-fold more ethanol (1.76 g/l) than cells composed of an equal amount of each cell type. Next, the fermentation performance of yeast consortium using steam-exploded CS as the sole carbon source was further investigated. The optimized cellulase-displaying consortium produced 20.4 g/l ethanol from 48.4 g cellulose per liter after 72 h in the presence of a small amount of cellulase reagent (0.9 FPU/ml), suggesting the feasibility of the cellulase-displaying yeast consortium for simultaneous saccharification and fermentation. Although several studies have been carried out on establishing a cell-displaying yeast consortium [25-27], few reports of direct ethanol fermentation from pretreated lignocellulosic material have been published. The combined cell system describ ed here could become the basis for the eventual direct ethanol production from insoluble cellulosic materials.

A Comparison of On the Road and Crying of Lot 49 Essay -- comparison c

In both Jack Kerouac’s, On the Road, and Thomas Pynchon’s Crying of Lot 49 the characters act in a deviant manner outside of social norms. This in turn leads to a deviant sub-cultural group which competes with the institutionalized authorities for power. Deviance in both novels is usually defined as a certain type of behaviour, such as an inebriated professor babbling on in a lecture hall filled with students or a group of teenagers frolicking naked in a city park on a hot and sunny afternoon. However, deviance can also encompass both ideas and attributes (Sagarin, 1975). The primary understanding of deviance rests in the reactions of observers, something becomes deviant because an individual, group or society takes offense and reacts negatively (Cohen, 1966, Lofland, 1969). These negative reactions occur because onlookers interpret what they see and hear as being bad, insane, strange, immoral, non-conforming, or wrong. Negative responses do more than define deviance; t hey serve as mechanisms of social control and power. In examining these novels from a sociological perspective, both Kerouac and Pynchon examine conflicts between mainstream society and sub-cultural groups. The deviant behaviour, thoughts, and attributes observed from the characters within the novels provide a strong argument for Austin Turk’s conflict theory of deviance, which examines power and cultural conflict as a basis for deviant behaviour. To begin, Austin Turk’s conflict theory of crime divides society into two groups: those with power "the authorities" and those without power "the subjects". In Pynchon’s novel The Crying Of Lot 49, this is realized by contrasting Pierce Inverarity, a California real estate mogul to those of low social economic class... ...sh-Catholic background, and his resemblance to a Fitzgerald hero, with a tragic death and foul dust floating in the wake of his dreams (153).    However, both of the novels express those subjects living by values beyond the social norms as having some power to change societal norms. Introduction The main purpose of the project scope statement is to have a basic understanding of the scope of a project, its deliverables, work required by deliverables to complete, and give common scope understanding to all stakeholders. Project Purpose and Justification The project of Great India Taste Restaurant has been approved by considering the demand of Indian food in the GTA by the related peoples. It has also been started for serving and creating a choice amongst the various types of world food in the Canadian market. Nowadays lots of immigrants are migrating here in Canada from India and South Asia. Therefore, the demand for their food is also increasing. Though there are some restaurants in GTA but they are not up to the mark. So, to be expert and perfect in every field of hospitality and service, we are here to work on this project. There is high probability of the success of this project as the public opinion has been considered. Scope description This project (restaurant) will be serving almost all the vegetarian and non vegetarian varieties of Indian food and related drinks and deserts such as Chana masala, Dal Makhani, Shahi Paneer, Butter chicken, Lassi, Naan, Puri and many more items. The specialty about all these food items would be that they will be prepared in clean and hygienic environment and made by the expert chefs. Due to increase in south Asian population in the selected area, this will not only keeps our restaurant busy but also may demand to open another chain of restaurant in the GTA. High Level Requirements In order to complete this project, we need to grant permission from Canadian Food Inspection Agency (CFIA) and other related bodies. For this we will have to ensure them the process of food preparation will be done by experts and in clean, hygienic environment and will have good nutrition value. Another high level requirement is that we have to construct our site as per the rules prescribed by the city and meet their requirements. These tasks are not difficult but they require high formalities and safety procedures which will ensure the success of the project. Project Boundaries As the name suggests, this project will serve only Indian food nothing out of that, no Chinese, no Italian, no Thai and so on. It will serve both vegetarian and non vegetarian food but it will not serve alcoholic drinks. It will charge affordable and fair price to the customers so to maintain goodwill and long term customer relationship. Project Strategy Number of strategies will be followed and experimented in this project. These are just for the promotion, dynamic environment, customer attention and retention and attracting more customers. This will increase our profitability and our competing power in the market. Deliverables Several deliverables will be produced as a result of successful completion of this project. The project will be considered successful only on the meeting of the following deliverables. These deliverables are: All the requirements described by various licensing bodies of Ontario must be met. It must be effective and efficient in food preparation and ensure better customer service It must generate enough profits to develop, maintain and improve services. Consider Thomas Hardy’s Essay

In the dictionary the word Isolation means to be separate or a part from others. I think if someone is isolated then it would be a negative aspect because they are alone and have nothing to do with anyone else. The reason for the various isolations in the two texts differs because of the different time periods they were set in. In the two books I am comparing is the â€Å"Withered Arm† by Thomas Hardy and â€Å"Of Mice and Men† by John Steinbeck. I feel that in both texts there are a few characters with their lives or they had illegitimate children. The text â€Å"Of mice and Men† was written in the 1930’s the time period of the depression. In the most of John Steinbeck’s books he describes the life of people working on the land and that his characters are all often poor. This was because he wanted to show how real the life was during that time. Within his books men and women rarely understood each other because of the lack of communication. During the time when John Steinbeck wrote â€Å"Of Mice and Men† people would frown upon coloured people, the handicapped, the old and women. They also were prejudice about women. The text the â€Å"Withered Arm† was written in the late 19th century in England. The book is set in a place called Wessex and the expectations at the time are different compared to the ones we have in today’s society. In that time social classes were expected to marry inside their class and not below that. Otherwise this would be considered a disgrace to the family of the higher class. Thomas Hardy introduces Rhode as his First character who is isolated. He establishes her isolation as her sitting on her away from the others â€Å"Where a thin fading women of thirty milked somewhat apart from the rest. † The word fading and somewhat apart from the rest reveals that she was separated from the rest of the milkmaids. Further on in the first chapter Hardy also establishes her loneliness by where she lives â€Å"†¦ lay part from that of the others, to a lonely spot high above the water-meads†¦ † this also show her isolation. The characters who are lonely are Candy, Crooks and Curley’s wife in the text â€Å"Of Mice and Men. † They are lonely because each one of them is different from everyone else. Such as Crooks is black, Candy is disabled and Curley’s wife is a women. The reason for them being lonely is because of the society’s attitude, which they lived in. The two characters that are not lonely are George and Lennie. This is as they have each other for company whereas everybody does not. Lennie and George treat each other, as they were family because they have been together for a very long time. â€Å"Because I got you an’___ An’ I got you. We got each other, that’s what, that gives a hot in hell about us’. † That quote indicates what Lennie say to George about them having each other to look after and keeping each other company. George needed Lennie because he wanted someone who would listen to him and not say anything. Lennie need George’s because he is not able to handle himself and also George promised Lennie’s aunt that he would look after him. Thomas Hardy describes Rhodes Brooks house as it old and worn † It was built of mud walls, the surface of which had been washed by many rains†¦ left none of the original flat face visible†¦ here and there in the thatch above a rafter showed like a bone protruding through the skin. † This suggests that Rhode did not a great house and that it was old just like her. The quote also indicates that the house had been through some hard times. In the text â€Å"Of Mice and Men† John Steinbeck writes about one female character that is known as Curley’s wife. As the reader I feel sympathy for her, as she is the only woman on the ranch. She is also isolated because her husband does not like her talking to any of the other workers, and also her own husband hardly speaks to her. The author has not given her a name because during that time period women were not as important as men, furthermore she is so isolated that the author decided on not given her a name. The effect this has on the reader is that she belongs to Curley unlike her being an individual person. So therefore the reader feels a little bit of sympathy for her. Thomas Hardy writes about a little boy in his book † Withered Arm† who is also nameless like Curley’s Wife. In the book he is referred as Rhode’s son. Rhode’s son is isolated because his mother had an affair with Farmer Lodge. The main reason that he is isolated is that his mother moved further away from the farm, so therefore he does not live close by to anybody. I feel that Rhode’s son is a victim of the circumstances between Farmer Lodge and Rhode because he was a product of their affair, and then Rhode moved away so she could be away from society. As a result Rhode’s son does not have friends. John Steinbeck writes about another isolated man named Crooks. The reason Crooks is so isolated is because during the time period everyone was prejudice against coloured people, and he was black. Another reason he is isolated as that he isolates himself to have some respect in the white world. Crooks was born with a disability, therefore he feels that he need to isolate himself further. He makes friends with Candy and Lennie as they isolated on the farm. When Crooks first meets Candy and Lennie it says † †¦ he kept is his distance and demanded that other people keep theirs. † This indicates that Crooks does not want anyone to be close to him. Though as the story goes on he makes friends with Candy and Lennie. I feel sorry for Crooks for being coloured and handicapped as he is very friendly person. In both the texts that I read there are two more handicapped people, named Gertrude from the † Withered Arm† and Candy from † Of Mice and Men†. Candy is isolated as he is old and has only got one arm since it was cut off when he was working on the farm. Gertrude isolated because one night Rhode cast a spell on her arm. In both texts Gertrude and Candy are alike because of their arms. They are not as isolated as Crooks or Rhode, since they only have a small isolation problem, unlike the rest. In the beginning of the text † Withered Arm† Farmer Lodge is presented as being bold and proud. He is proud as he has a new wife Gertrude, who is a lot younger than him and plus she is very beautiful. Gertrude starts to get a problem in one of her arms, as time passes her arm gets worse, therefore Farmer Lodge feels as she is not as pretty as before. This makes Farmer Lodge change. He also feels depressed since his son is hung and this changes him. The reason for him to isolate himself is that all these problems are occurring around him and he feels that it is his fault. So he moves away from his farm. In conclusion I consider that isolation is a negative aspect in someone’s life. I think that when people are isolated dreaming keeps them from getting trough. Like for instance Lennie and George’s dream to open up a farm or their own and Rhode’s dream to get back with Farmer Lodge. Work force Up-skilling

Organisations usually view its work force as their driving force. The work force is believed to be the determinant of how far the company will go in terms of achieving its objectives. Organisations usually formulate strategies aimed at upskilling this workforce. This is usually done through methods aimed at improving the skill possessed by its workers. Organisations usually formulate strategies, which are aimed at achieving their workforce level objectives (Argyris, 1976p. 34-56). Since the organisations are required to try and create a team that possess high skills the process of coming up with a workforce of higher skill need not be universally accepted. The companies are supposed to adopt a way of moving towards higher skill that fits its requirements (Terry, 1960p. 104-109). This method of creating a workforce of higher skill should be formulated the best way possible an organisation can manage considering its requirement in terms of organizational strategy (Torbert, W. 2004p. 41-42). Workforce structure. Work force in organisations is usually composed of two categories depending on the type of workers. These categories are: – (i) Skilled workers (ii) Unskilled workers Skilled workers refers to workers of organisations who are believed to possess qualities such as possession of a certain level of good education in their field of specialisation. The Skilled workers are also believed to have acquired experience in their fields of specialisation. Skilled workers are required to have other features that enable them to be termed as efficient in their duties. Examples of these features include good communication skills and also interpersonal skills which aid incase one has to work in a group. Skilled workers are also termed as having broad level of general knowledge which is a factor in efficiency in their duties. Examples of skilled workers include Mechanical engineers, doctors, management accountants, finance officers and also civil engineers. Unskilled workers are workers in organisations who do not need to have any educational (formal) background. These workers need not have any experience to get jobs in an organisation. Unskilled workers are believed to have no skill for the jobs they carry out for organisations. These are the workers in an organisation who are believed to be at the bottom under the consideration of the level of qualifications. Most of the employees who are unskilled in organisations include cleaners and also loaders in stores. According to statistical data in the UK the skilled workers usually get the task of seeking employment easy. This is because most of the employment vacancies available are skilled occupations. It has been found that out of every 1,000 skilled job seekers, 75% end up succeeding. The statistical data has also revealed that the organisations in the UK give better working conditions to the skilled employees. It has been found that this preference in terms of better working conditions to skilled workers is usually an attempt by the organisation to maintain skilled teams in their organisation. This is not the case with unskilled workers. These workers find it hard to get good working conditions in organisations (Bass, & Avolio1995p. 100-123). The reason deducted for this occurrence is the fact that unskilled workers are readily available and incase their turnover tends to be high, there is no actual loss to the organisation. Human resource management refers to the process of handling matters concerning work force in an organisation. Each organisation usually has a department overseeing the efficiency of its work force. The human resource management is done by application of various models such as the goal Management Model. This human resource management model emphasizes on the need for organisations to handle their human resource in a manner likely to lead to the goal achievement. In the even of workforce upskilling a company should create a team of higher skill through assessment of the company’s goals. The organisations should use strategies likely to incorporate the needs of the organisation in terms of employee level of skills. The organisation should apply a strategy based on the level of skills required in achieving given goals. Another example of these models is the Culture change Model this is a model in human resource management which emphasizes that organisation changes their culture (House, 2004p. 23-31), and therefore the employees / workers should be upskilled to match the changes. Kurt Levin developed this model. Open systems Model is another model, which emphasizes that the human resource in an organisation should be managed as per the open system concept. This concept views systems as units that exchange requirements. The model argues that in employee upskilling the organisations should be able to get new employees from outside but not just develop the existing workers (Lowman, 2002p. 54-61). This model gives an organization option of acquiring workers of higher skills. All the three models described above show that there is no generally accepted way of upskilling an organisation workforce. The models suggest that an organisation should adopt a movement, which will be in agreement with the events at the organisation. For example when the organisation is faced with a change in its organizational culture, Lewin’s human resource management model should be adopted. When an organisation finds out that it cannot get the level of skill in the current workforce the open system human resource management model should be adopted, or just develop the existing workers to the level required, a characteristic of the Path Goal leadership model. Organisations usually emphasise on employee development in the attempt to reach a higher skilled work force. Employee development refers to the process in which employees of an organisation are involved in training in the organisation. Employee development involves a programme whereby the employees participate in their own will, the employees schedule their learning hours, the content to be learned is usually extra of what the job training requires and the workers select the content to learn without the influence of the organisations. Most companies in the UK emphasize on employee development since it is believed to give higher skilled employee workforce. Some of the companies in the UK, which emphasize on employee development, include Siemens UK and also Roll Royce Cars. These companies give a given level of provision in terms of sponsorship for employee training in the employee. The personal development theory in employee development argues that workers should be in a position to enhance the programme of employee development. The workers are required to be willing and interested in developing as employees of a given organisation. The employee motivation theory argues that the organisations should put efforts in the process of employee development. The organisations efforts should be in the form of programmes likely to boost the morale of employees (Stacey, 1992p. 31-39). The Benziger theory also provides directions on the employee development programmes in an organisation. In the process of employee development the organisations can create programmes bearing in mind that workforce usually consist of skilled and unskilled workers. The organisations should create programmes likely to give formal education, experience, skills and also general knowledge to the unskilled workforce. This is likely going to improve the skill of employee team hence likelihood of a success towards organizational strategy. The organisations should create programmes which will enable the skilled workforce improve their efficiency and also make them acquire certain level of vital attributes such as confidence and self-initiative (Warneka, 2006p. 23-29). Employee development should be structured in a way likely to make the organizational strategy give desired results. The employee development should be done in a way likely to put all the levels of the work force at a higher level of skill (Burns, 1978p. 65-71). Organisations should not copy the programmes adopted by their competitors or other comparable organisations. Copying of employee development programmes would not help organisations in the process of up-skilling its workforce since it might not suit the organisation. The organisations should structure these programmes as per their workforce requirement bearing in mind that the best workforce is that which is at a higher level in terms of suitability to a specific organization. Conclusion. Organizations should try as much as possible to avoid the perception that, workforce up-skilling is standardised (Heifetz, 1994p. 12-19). Organizations should formulate organizational strategies aimed at effective teams’ suited to the needs of specific company.
